In Vedic Astrology the Shashthamsa is not used. This is the Dvisional Chart 6. Let's think through why it is an important divisional chart.
(1) It deals with enemies. This is the only chart where it shows the interaction of the individual with society on the initial basis of negative relationships (unlike D-10 (work and actions with society), D-30 (evils arising from one self and not interaction) etc.)
(2) It deals with health.
(3) It deals with the expanded concepts found in our 6th House of the Rasi including litigation (more so that the D8 although D8 must also be considered) etc.
Of the three Dusthana Houses in the Rasi, only the 6th House has a eleventh-third house relationship with the 8th house which is also a Dusthana. What this means is that if the 6th House is strong (the enemies), it would contribute to the fall of the native ie. contribute to the strength of the 8th House. Thus two Dusthana Houses are automatically involved if one is dealing with the 6th House even though it may be the least powerful amongst the three Dusthana Houses.
Thus the Shashthamsa Karya Houses are the 6th and the 8th. And of course its trine houses to the 6th would be the ones to counteract its evil effects ie. the 2nd and the 10th in the D6. The 10th to the 6th ie. the 3rd which involves actions and writing will set things straight also to avert the consequences of the 6th House.
Let's start with my D6 chart.
I have Jupiter in Cancer in 10th, Moon in 6th, Sun in 8th, Venus in 3rd with a Libra Ascendant.
My ascendant ruled by Venus in 3rd shows that I always try to avert enemity by forseeing any potential dangerous situation (in philosophy they call it the "slippery slope") for me which would put me in a jam and I take action through telling the person in a nice way or even emailing or writing to him
Moon in 6th (and being the Lagna Lord and the Atmakaraka) shows I would always encounter enemity in public life (the Moon). Sun in Taurus in 8th shows that enemity that piles up on me may even end up in their taking concerted action against me when the opportune time arises (phew! luckily I am not in politics).
But Jupiter has a good reputation and is exalted in Cancer in the 10th. It disposits Venus an the Moon. With a stroke of luck all enemity vanquished and hence the chain of the 8th to the 6th falls as well (remember what I menioned earlier about the eleventh-third house relationship between the 6th and the 8th). Everytime I entered Jupiter sub-dasa period (besides Mars and Ketu), problems get solved.
More research would have to be done on the D6 chart.

I would be very curious about what my D6 chart says as I have a very emphasized 6-12 house axis to start with. I think of myself as a 6th house person and rather enjoy dealing with challenges. My health is good, too. Data: Feb 17 1955, 6pm, Mumbai. It might be instructive to look at the positive side of the this house (work, money).

What happens if there is both a strong planet and a weak one? What I am trying to figure out is: Is it good to have a weak sixth lord and a strong planet posited there? Or, should both be weak ... to show that the enemies (external events?) cannot hurt?

Although this question was not directed to me, although I will like to add my two cents.
D6 chart shows, troubles, enemies and diseases. as explained by Respected Leong.
6L if placed in 3H, 6h, 8H,12H and 11H is good if strong. for overcoming enemies. But if 6L placed in 8H may not be good for Health and may indicate some serious diseases.
While a strong malefic planet in 6H is good for overcoming enemies such people are courageous and energetic enough to fight and win over enemies. while a strong benefic in 6H is not good for overcoming enemies. such people are usually not interested in fighting and even if indulged in a fight they tend to leave the fight halfway. Although this is only a thumb rule and other factors need to be studied properly before reaching a conclusion
