Dear Rita,
ISHTA KAAL IS POPULARLY KNOWN AS ISHTA GHATI. Istakaal is the difference between sunrise and time of birth. HERE THE BIRTH TIME MUST BE CONVERTED TO LOCAL MEAN TIME.
This is used in BTR. YOU CAN GOOGLE IT ABOUT THE PROCEDURE

Do I need to change any settings in JHora to derive the correct value of Janma Ghati? As Ramananji says it has to be Local Mean Time, how do I ensure JHora is using that?

However I tried to search on the internet, but the examples I found are not very clear to follow. I would be grateful if you could use this time of birth as an example and explain how to calculate Ishta Kaal:

Dear Rita,
The sun rise time given in j hora for the date of birth given is....6.31.50...Your time of birth is 16.30.All we have to do is just find out as to how many ghatis have elapsed between the the sun rise and the time of birth.One ghati is 24 minutes.
To make calculations easy...Let us assume the sun rise time to be 6.30 AM...The birth time is 16.30 hours. so 10 hrs has elapsed and 10 hrs is 25 ghatis. , as 2.5 ghatis is one hour.
I think j hora calculates this for you and you need not take the pains to convert an hour in to ghatis and ghatis in to vighatis.

JHora gives you an option to use LMT (local mean time) for chart generation. There is a check box in the birth data dialogue. If you are calculating manually as Ramananji has suggested you can use the local sun rise time published in a regional almanac (panchaang) and the birth time as per local standard time.
